Job Description

Job Summary

The Scadden laboratory is dedicated to discovering the principles governing blood cell production, with the ultimate goal of guiding the development of therapies for blood disorders and cancer. The Lab is seeking an experienced scientist to provide scientific and administrative leadership for three projects. The Research scientist will collaborate with the Principal Investigator to develop and execute complex and specialized research studies as part of an industry-sponsored collaboration. They will also collaborate with and manage other scientists recruited to work on the project.

Position Description

Independently lead and execute large-scale small molecule screens for hit identification and contribute to follow-up experiments throughout medicinal chemistry and lead optimization.

  • Lead the development of assays for small molecule discovery
  • Miniaturize and adapt cell-based and biochemical assays to automated screening systems
  • Propose and develop innovative approaches to small molecule discovery based on the latest technologies and publications in the field
  • Analyze, critically interpret and present data to industry partners, and recommend courses of action based on datasets and discussions with biologists and chemists.

Work with multi-scientist team to develop subcutaneous organoid with appropriate signaling molecules to improve t-cell reconstitution.

  • Develop protocols to achieve reproducible organoids for large scale work in mice, humanized mice, and higher order species.
  • Develop protocols to analyze T-cell recovery in higher order species
  • Explore additional molecules to improve stem cell content in subcutaneous organoids to serve as a means to improve gene therapy induction in HSCs.

Basic Qualifications

MS in Biological/ Pharmaceutical Sciences or a related field with at least 7 years of hands-on experience in academic or pharmaceutical/biotechnology labs.

Additional Qualifications and Skills

  • Experience in molecular biology techniques and deep understanding of normal and malignant hematopoietic biology are required.
  • RNA/DNA extraction/purification and manipulation (e.g. enzyme digestion and modification, agarose gel electrophoresis, qPCR, RT-PCR)
  • Taqman and gene expression analysis
  • Flow Cytometry, SDS-PAGE and Western blotting
  • Mammalian cell culture, genetic target modulation (siRNA, shRNA, CRISPR) via cell transfection and sterile technique
  • Hematopoietic stem cell isolation and culture techniques.
  • Lentivirus and adeno-associated virus production and cell transduction.
  • Experience developing and optimizing enzymatic biochemical assays.
  • Experience working with bone marrow transplants in mice is required.
  • Good understanding of T-cell lineage and associated assays including TREC.
  • Bone tissue image analysis ? microCT and histology/morphology
  • Experience with high-throughput robotic platforms is a plus.
  • Knowledge of the latest technologies and novel approaches to small-molecule drug discovery
